This study investigated whether SHP2 activity is involved in cell proliferation and activation of the JAK2-STAT3-SOCS3 pathway in human prostate cancer (PCa) cells. Expression of SHP2 was determined in PCa cells and human normal prostate tissues. SHP2 activity was determined by the dephosphorylation of a phosphopeptide, pY317-SHP2 substrate. Cell proliferation was measured using the MTT assay. The expression levels of several proteins, including SHP2, pY317-SHP2, pSTAT3, SOCS3, Bcl-xL and survivin were detected by Western blotting. The involvement of SHP2 in the activation of the JAK2-STAT3-SOCS3 pathway was examined using specific siRNA. We found that SHP2 expression was up-regulated in PCa cells and tissues. In PCa cells, SHP2 activity was lower than in normal human prostate epithelial cells. Knockdown of SHP2 decreased proliferation and reduced the expression of pY317-SHP2 and pSTAT3. In addition, knockdown of SHP2 enhanced the expression of SOCS3, and knockdown of SOCS3 restored the decreased proliferation induced by SHP2 knockdown. Moreover, we found that knockdown of SHP2 reduced Bcl-xL expression, but increased survivin expression. In contrast, knockdown of SOCS3 reversed the effects of SHP2 knockdown on the expression of Bcl-xL and survivin.
